چکیده

Homoaconitic acid, the second intermediate of the proposed pathway for lysine biosynthesis in yeast, is accumulated in the growth medium of a lysine-requiring mutant. This acid has been identified on paper and column chromatography by comparing it with authentic cis-homoaconitic acid. The infrared spectrum of the isolated material was identical with that of synthetic cis-homoaconitic acid. In addition, the chemical structure of the enzymatic product has been verified by degradation to glyoxylic and alpha-ketoglutaric acids after treatment with KMnO(4) and HIO(4) and by catalytic reduction to the saturated acid 1,2,4-butanetricarboxylic acid. The isolated homoaconitic acid was also identified as a substrate for a purified enzyme preparation of homoaconitase.

Accumulation of tricarboxylic acids related to lysine biosynthesis in a yeast mutant.

Accumulation of tricarboxylic acids related to lysine biosynthesis has been demonstrated in a yeast mutant, Ly,,, which requires lysine as a growth factor in synthetic medium but can substitute a-aminoadipic acid for the nutritional requirement of lysine. Both radioactive and nonradioactive intermediates have been isolated from the growth medium under appropriate conditions and have been identi...

Homocitric acid accumulation by a lysine-requiring yeast mutant.

The accumulation of fi-carboxy-P-hydroxyadipic acid (homocitric acid) in the culture medium of the lysine-requiring yeast mutant, Ly4, grown on limiting amounts of lysine, has been demonstrated. Identity of the biologically formed acid with synthetic homocitric acid was established by comparison of infrared spectra, RF values from paper chromatograms, optical rotations, and melting points. Radi...

Biosynthesis of Lysine from a-Ketoadipic Acid and oa-Aminoadipic Acid in Yeast

Studies of the nutrition of certain lysine requiring mold mutants indicate that L-a-aminoadipic acid (AAA) or a-ketoadipic acid (KAA) are precursors of lysine. For example, Mitchell and Houlahan (1948) found that certain lysine requiring mutants of Neurospora crassa would respond to either lysine or AAA, and Windsor (1951) subsequently demonstrated that f-C'4-a-aminoadipic acid was completely i...

A lysine accumulation phenotype of ScIpk2Delta mutant yeast is rescued by Solanum tuberosum inositol phosphate multikinase.

Inositol phosphates and the enzymes that interconvert them are key regulators of diverse cellular processes including the transcriptional machinery of arginine synthesis [York (2006) Biochim. Biophys. Acta 1761, 552-559]. Despite considerable interest and debate surrounding the role of Saccharomyces cerevisiae inositol polyphosphate kinase (ScIPK2, ARG82, ARGRIII) and its inositol polyphosphate...
